What is magnesium glycinate?
Magnesium glycinate is a form in which magnesium is bound (or “chelated”) to the amino acid glycine.
You may also see the name magnesium bisglycinate. The terms are often used interchangeably in consumer products, but product names alone do not tell you the precise raw material, hydration state or final formulation.
That distinction matters because a front label can state a large amount of magnesium glycinate while providing a much smaller amount of elemental magnesium. The nutrition or medicine panel is the more useful place to look.
One more thing about this form. Glycine is not simply a carrier. It is an inhibitory neurotransmitter in its own right, and it has been studied on its own for sleep, with a proposed mechanism involving a drop in core body temperature, which is one of the physiological cues for sleep onset. That is part of why this particular combination attracts interest for evening use.
The dose is where it gets interesting. Those glycine studies used around 3 g. A supplement providing 100 mg of elemental magnesium as bisglycinate carries roughly 0.6 g of glycine, and one providing 250 mg carries roughly 1.5 g. So a typical serve sits below the amount that has been studied, though not by as much as you might assume.
This cuts in two directions, and both are worth holding. It is a reasonable argument for why the combination might suit the evening. It is also a reason to be careful reading the 2025 sleep trial, which used 250 mg of elemental magnesium and therefore delivered about 1.5 g of glycine alongside it. That trial tested a compound, not magnesium by itself, and its result cannot be cleanly attributed to the mineral.
The glycine sleep research is also thinner than it first appears. The trials were small and relied on subjective sleep measures, and much of the early work came from researchers at a company that manufactures amino acids. More recent independent reviews describe the evidence as promising and call for properly powered randomised trials. Worth knowing, not worth building a purchase decision on.

02 · Reading the label
Start with elemental magnesium.
Elemental magnesium is the actual quantity of magnesium contributed by the ingredient, not the total weight of the magnesium compound.
Every magnesium supplement is a compound: magnesium joined to something else. That partner has weight too, and on most labels it makes up the larger share. Magnesium is about 14% of the weight of magnesium bisglycinate, so 1,000 mg of the compound supplies roughly 141 mg of magnesium.
The proportion varies a great deal between forms. Magnesium oxide is around 60% magnesium by weight, and magnesium citrate around 16%. A physically smaller oxide tablet can therefore carry more magnesium than a larger glycinate capsule, which is why comparing the front-of-pack number across two different forms tells you very little on its own.
Weight percentage is not the whole story either. How readily a compound dissolves, how well it is tolerated and how the product is formulated all affect what a serve actually does. Reviews of magnesium bioavailability generally report better absorption from organic magnesium salts than from oxide, while noting that trial designs vary widely and that individual factors matter.
On an Australian label, look past the front of the pack to the medicine panel. You want the line that reads “equiv. magnesium”, or a formulation such as “magnesium (as magnesium bisglycinate)” followed by an amount. That figure is the elemental amount, and it is the only number that lets you compare two products honestly.
One caution on using recommended dietary intakes as a target. Australian RDIs cover total magnesium from all sources, food included, and most people get the bulk of theirs from food. An RDI is a reference point for total intake, not a supplement dose.

03 · The research
What research tells us so far.
Magnesium's role in the body is well established, and research into this particular form is building. Here is what has been shown in people so far.
It helps to separate two questions that often get blurred together. What does magnesium do in the body, and what has this particular form been shown to do in people?
The first is well established. Magnesium is an essential mineral that acts as a cofactor in more than 300 enzyme systems, supporting muscle and nerve function and the formation of protein, bone and DNA. None of that is in dispute.
The second is a smaller and much newer body of work. Most magnesium trials have used other forms, or have not specified the form at all, so a finding about magnesium in general cannot be read as a finding about magnesium glycinate. We keep those two things apart throughout this site.
Four human trials have used this form specifically. A 1994 crossover study in 12 patients who had undergone ileal resection compared magnesium diglycinate with magnesium oxide using a stable isotope. Total absorption was similar across the group as a whole, magnesium from the chelate appeared in the blood earlier, and it was better tolerated by every patient. A 2015 trial in 80 pregnant women found that magnesium bisglycinate reduced both the frequency and the intensity of leg cramps more than placebo over four weeks. A 2024 crossover study in 40 adults compared bisglycinate, citrate and oxide and found a similar total rise in blood magnesium across all three over six hours. And a 2025 randomised trial in 155 adults reporting poor sleep found a statistically significant improvement in insomnia severity after four weeks, a result the researchers themselves described as modest.
Taken together that is a promising but early picture. The trials are small, most run for four weeks or less, and two of the four used specific populations rather than the general adult public. A 2026 systematic review of twelve magnesium sleep trials reached a similar conclusion: modest benefits on some self-reported measures, and a clear need for larger and longer studies. That is a normal place for research on a specific compound to sit, and it is more than many supplement ingredients can point to.

This randomised, double-blind study is the first to test this form for sleep, and the result on its main measure was positive. The authors described the improvement as modest, noted that the outcomes were self-reported, and recommended longer trials using objective sleep assessment to build on the finding. The trial used 250 mg of elemental magnesium as bisglycinate, which also supplies roughly 1.5 g of glycine, so the result belongs to the compound rather than to magnesium alone.
04 · Context
How does it compare with other forms?
A useful comparison separates what a compound is from what has been demonstrated in people.
Three separate things get compared when people discuss magnesium forms, and they are often run together: how much magnesium the compound contains by weight, how well that magnesium is absorbed, and how comfortable it is to take.
On the first, oxide wins easily, at around 60% magnesium by weight against roughly 14% for bisglycinate. On the second, the evidence is thinner than the marketing around it suggests. The 2024 crossover study in 40 adults found a similar total rise in blood magnesium from bisglycinate, citrate and oxide over six hours. The 1994 study found similar total absorption between the chelate and oxide across its group, with a clear advantage appearing only in the four patients whose absorption was most impaired to begin with.
On tolerability the picture is more consistent. The 1994 study reported better tolerance of the chelate in every patient it enrolled. In the 2025 sleep trial, 93% of participants reported no adverse events, and side effects were less frequent on magnesium bisglycinate than on placebo. Citrate, by contrast, is used as an osmotic laxative at higher doses, which tells you something about its effect on the gut.
So the honest summary is this. Form is one variable among several, and the elemental dose you actually take matters at least as much as which compound delivers it. Where glycinate has the clearest support is comfort rather than dramatically superior absorption, and comfort is not a trivial thing for something taken daily. Anyone claiming a large absorption advantage for one form over another is ahead of the current evidence.

05 · Safety
Use it thoughtfully.
The right guidance helps make supplementation safer, clearer and better suited to your individual circumstances.
Magnesium supplements are generally well tolerated, and the most common problems are digestive: loose stools, nausea and abdominal cramping. These are dose related, which is part of why the amount you take deserves more attention than the form you choose.
There is a tolerable upper intake level of 350 mg per day for supplemental magnesium in adults. That limit applies to magnesium from supplements and medicines only. It does not apply to magnesium from food, because healthy kidneys clear dietary excess efficiently.
Kidney function is the thing to take seriously here. Magnesium is cleared renally, so reduced kidney function is the main circumstance in which supplemental magnesium can build up to harmful levels. That is a conversation with a doctor, not something to work out from a website.
Magnesium can also interfere with the absorption of several common medicines, including some antibiotics in the tetracycline and quinolone families, bisphosphonates, and levothyroxine. Separating the doses by a few hours usually resolves it, but the right interval depends on the specific medicine, and your pharmacist can check that for you.
One last thing worth saying plainly. Blood magnesium tests reflect body stores poorly, because most of the body’s magnesium sits in bone and inside cells rather than in the blood. A normal reading does not rule out low stores, and a supplement is not a way to investigate symptoms. If something feels wrong, that deserves a proper look rather than a guess.
Speak with a doctor or pharmacist before use if:
